MMAE and MMAF are potent tubulin-inhibiting payloads used in antibody-drug conjugates (ADCs). Both disrupt microtubules, halting cell division and inducing apoptosis in targeted cancer cells.
Their key difference lies in membrane permeability. MMAE is membrane-permeable, allowing a potent “bystander effect” to kill neighboring tumor cells, ideal for heterogeneous cancers. MMAF, due to a charged C-terminal group, is not membrane-permeable. This confines its lethal activity strictly to the antigen-positive cell that internalizes the ADC, offering a more precise attack suitable for targets expressed on healthy tissues.
