Monomethyl auristatin E (MMAE) is a potent synthetic derivative of dolastatin 10, a natural peptide originally isolated from a marine sea hare. It functions as a microtubule-disrupting cytotoxic agent. MMAE works by binding to tubulin with high affinity, inhibiting its polymerization into microtubules. This disruption halts cell division during mitosis, ultimately triggering programmed cell death (apoptosis) in rapidly dividing cells.
A key characteristic of MMAE is its exceptional potency; it is effective at picomolar to nanomolar concentrations. However, its high systemic toxicity prevents its use as a standalone chemotherapy drug. Instead, MMAE has become a cornerstone payload in the field of antibody-drug conjugates (ADCs). In ADCs, MMAE is covalently linked via a stable, cleavable linker to a tumor-targeting monoclonal antibody. This design allows for the selective delivery of the toxic payload directly to cancer cells expressing the specific antigen, thereby minimizing off-target effects and broadening the therapeutic window.
