Monomethyl Auristatin F (MMAF) is a potent anti-tubulin agent used as a payload in antibody-drug conjugates (ADCs). It inhibits microtubule assembly, arresting cell division and inducing apoptosis.
A key distinction from the similar MMAE is its charged C-terminal group, which prevents efficient cell membrane diffusion. This eliminates the “bystander effect,” ensuring cytotoxicity is confined strictly to antigen-positive cells that internalize the ADC. This feature makes MMAF particularly suitable for targeting solid tumors where precision is critical.
