CD19 is a 95 kDa transmembrane glycoprotein that serves as a critical surface marker and functional regulator of B cells. It features two extracellular C2-type immunoglobulin-like domains and a broad cytoplasmic region involved in intracellular signaling. Expressed throughout B-cell development—from early progenitors to mature B cells—CD19 is essential for B-cell activation, proliferation, and differentiation into plasma cells. On the cell surface, it forms a co-receptor complex with CD21 and CD22, modulating B-cell receptor (BCR) signaling thresholds and enhancing cellular responses to antigens. Due to its B-cell–restricted expression and pivotal role in B-cell function, CD19 has become a key therapeutic target in hematologic malignancies, particularly in chimeric antigen receptor (CAR) T-cell therapies for B-cell acute lymphoblastic leukemia (B-ALL) and non-Hodgkin lymphoma. Its involvement in autoimmune disorders further underscores its immunological and clinical significance.
