CD25, also known as IL-2 receptor α chain (IL-2Rα), is a key component of the interleukin-2 receptor (IL-2R) complex. It is primarily expressed on activated T cells, regulatory T cells (Tregs), and some dendritic cells. As a low-affinity receptor for IL-2, CD25 alone binds the cytokine with weak affinity, but upon ligand engagement, it facilitates the recruitment of CD122 (IL-2Rβ) and CD132 (IL-2Rγ) to form a high-affinity ternary receptor complex. This structural reorganization enhances IL-2 signaling, which is essential for T-cell proliferation, differentiation, and immune regulation. In clinical contexts, CD25 serves as a biomarker for activated T cells and Tregs, and is a target in therapies for autoimmune diseases and certain cancers.
