CD3 is a multi-subunit transmembrane complex composed of five polypeptide chains: CD3γ, CD3δ, CD3ε, CD3ζ, and CD3η. These chains associate non-covalently to form heterodimers (γε and δε) and homodimers (ζζ or ζη), which together with the T-cell receptor (TCR) constitute the TCR/CD3 complex on the surface of all mature T cells. Each of the CD3γ, δ, and ε chains contains an immunoreceptor tyrosine-based activation motif (ITAM) in its cytoplasmic domain. Upon TCR engagement with MHC-presented antigen, ITAMs undergo phosphorylation, initiating intracellular signaling cascades that lead to T-cell activation, proliferation, and effector function.

CD3 plays an indispensable role in transducing antigen-specific signals from the TCR into the cell. In research and clinical applications, antibodies targeting CD3ε are widely used to activate T cells in vitro and are also harnessed in therapeutic contexts, such as in the development of bispecific T-cell engagers (BiTEs) for cancer immunotherapy. As a universal marker for T cells, CD3 is essential for immunophenotyping, functional studies, and therapeutic modulation of T-cell-mediated immunity.
