CD43 is a heavily sialylated transmembrane glycoprotein that plays a role in T-cell activation and immune regulation. Structurally, it consists of a mucin-like extracellular domain rich in serine and threonine residues, which supports extensive O-glycosylation and contains a potential N-glycosylation site; a transmembrane region; and a cytoplasmic tail harboring putative phosphorylation sites involved in signal transduction.
CD43 is expressed on the surface of thymocytes, T lymphocytes, monocytes, granulocytes, and certain B-cell subsets. It modulates cell adhesion, migration, and antigen-specific activation of T cells. Upon immune stimulation, CD43 can undergo ectodomain shedding—proteolytic cleavage that releases its soluble extracellular fragment into the microenvironment, a process thought to regulate leukocyte function and inflammatory responses.
While its precise signaling mechanisms remain under investigation, CD43 is recognized as an important immunoregulatory molecule with implications in autoimmunity, infection, and cancer biology. Its expression pattern and post-translational modifications make it a subject of continued interest in both basic immunology and translational research.
