CD45, also known as Leukocyte Common Antigen (LCA), is a transmembrane glycoprotein universally expressed on the surface of all nucleated hematopoietic cells. It functions as a key regulator of immune cell signaling through its intracellular protein tyrosine phosphatase (PTPase) activity, which modulates the activation of Src-family kinases such as Lck and Fyn in T and B lymphocytes. Structurally, CD45 consists of a heavily glycosylated extracellular domain, a single-pass transmembrane region, and a cytoplasmic tail containing two PTPase domains. Alternative splicing of exons encoding its extracellular region generates multiple isoforms, including CD45RA, CD45RB, CD45RC, and CD45RO, which are differentially expressed during leukocyte development and activation. For example, CD45RA marks naïve T cells, while CD45RO is associated with activated and memory T cells. Due to its pan-leukocyte expression and isoform-specific patterns, CD45 serves as an essential marker for immunophenotyping in research, diagnostics, and flow cytometry, and plays a critical role in both adaptive and innate immune responses.
