CD8 is a cell surface glycoprotein expressed primarily on cytotoxic T lymphocytes (CTLs). It functions as a coreceptor for the T cell receptor (TCR), assisting in the recognition of antigen presented by major histocompatibility complex class I (MHC-I) molecules. Structurally, CD8 exists as either a homodimer (αα) or heterodimer (αβ), with both chains belonging to the immunoglobulin superfamily. The extracellular domain of the CD8α chain interacts with the α3 domain of MHC-I, stabilizing the TCR–MHC-I interaction and enhancing T cell activation. Intracellularly, CD8 recruits the tyrosine kinase p56lck, which phosphorylates ITAM motifs on the CD3 complex, initiating downstream signaling pathways essential for T cell proliferation, differentiation, and cytotoxic function. CD8 is thus a critical molecule in adaptive immunity, enabling CTLs to identify and eliminate infected or malignant cells.
