HOX40L (also known as TNFRSF4 ligand or CD252) is a type II transmembrane protein belonging to the tumor necrosis factor (TNF) superfamily. It is primarily expressed on antigen-presenting cells such as dendritic cells, B cells, and macrophages. As the ligand for OX40 (CD134, TNFRSF4), HOX40L plays a critical role in T-cell costimulation and immune regulation. Upon binding to OX40 on activated T cells, it enhances T-cell proliferation, survival, cytokine production, and the formation of memory T cells. This interaction is particularly important for sustaining effector and memory T-cell responses in infections, antitumor immunity, and autoimmune inflammation. Given its function in modulating adaptive immunity, HOX40L has emerged as a promising target in immunotherapy, with ongoing research focused on agonistic or antagonistic strategies to enhance antitumor responses or treat autoimmune and inflammatory disorders.
