IFN-γ (Interferon-gamma) is a dimeric cytokine primarily produced by activated T cells (Th1, CD8⁺ T cells) and natural killer (NK) cells. It serves as a key mediator of innate and adaptive immunity, promoting macrophage activation, antigen presentation via upregulation of MHC class I and II molecules, and Th1-type immune responses. IFN-γ enhances antimicrobial and antitumor activity by inducing nitric oxide production, modulating cell proliferation and apoptosis, and coordinating immune cell recruitment and function.
Clinically, IFN-γ plays a dual role in immunity and pathology. It is essential for host defense against intracellular pathogens (e.g., mycobacteria) and viruses, yet its dysregulation is linked to autoimmune diseases (e.g., rheumatoid arthritis, inflammatory bowel disease) and chronic inflammation. In oncology, recombinant IFN-γ has been explored as an immunotherapeutic agent to enhance antitumor responses, and its expression levels are often assessed as a biomarker of immune activation in cancer and infectious disease monitoring.
