IL-12 (Interleukin-12) is a heterodimeric cytokine composed of two subunits, p35 (IL-12α) and p40 (IL-12β), primarily produced by antigen-presenting cells such as dendritic cells, macrophages, and B cells. It serves as a critical bridge between innate and adaptive immunity by promoting the differentiation of naïve T cells into T helper 1 (Th1) cells, enhancing interferon-gamma (IFN-γ) production, and boosting the cytotoxic activity of natural killer (NK) and CD8⁺ T cells.
Due to its potent role in driving cellular immune responses, IL-12 has been extensively studied in the context of infectious diseases, cancer immunotherapy, and vaccine development. It enhances antitumor immunity by stimulating IFN-γ-dependent pathways and improving antigen presentation. Clinically, recombinant IL-12 has been explored as an immunotherapeutic agent, though its systemic use is limited by toxicity. Current strategies focus on localized delivery or IL-12-based gene therapies to harness its immune-stimulating potential while minimizing adverse effects. Dysregulation of IL-12 is also implicated in autoimmune and inflammatory disorders, highlighting its dual role in immune activation and pathology.
