IL-13 (Interleukin-13) is a cytokine primarily produced by activated Th2 cells, group 2 innate lymphoid cells (ILC2s), mast cells, and basophils. It shares functional overlap with IL-4 and signals through a receptor complex involving IL-13Rα1 and IL-4Rα, driving key aspects of type 2 immune responses. IL-13 promotes B-cell class switching to IgE, mucus hypersecretion, airway hyperresponsiveness, and alternative activation of macrophages (M2 polarization).

Clinically, IL-13 is a central mediator in allergic diseases such as asthma, atopic dermatitis, and chronic rhinosinusitis with nasal polyps. Its role in tissue remodeling and fibrosis also implicates it in conditions like idiopathic pulmonary fibrosis and scleroderma. Therapeutic monoclonal antibodies targeting IL-13 have been developed for asthma and atopic dermatitis, underscoring its importance as a therapeutic target in type 2 inflammation and allergic pathology.
