IL-6 (Interleukin-6) is a key pleiotropic cytokine that regulates immune responses, inflammation, hematopoiesis, and the acute-phase reaction. It is produced by various cells upon infection or injury, promoting B- and T-cell activation, Th17 differentiation, and hepatic synthesis of acute-phase proteins like CRP. As an early and sensitive biomarker of infection, IL-6 levels rise rapidly but lack specificity, as elevations also occur in autoimmune diseases, cancer, and chronic inflammation. Dysregulated IL-6 signaling is implicated in conditions such as rheumatoid arthritis, cytokine storm, and multiple myeloma, making it a major therapeutic target. Antibodies against IL-6 or its receptor are clinically used to modulate excessive immune activation.
