TNF-α (Tumor Necrosis Factor-alpha) is a pleiotropic pro-inflammatory cytokine primarily produced by activated macrophages, T cells, and natural killer (NK) cells. It plays a central role in regulating immune responses, inflammation, and cell survival by binding to its receptors, TNFR1 and TNFR2, which activate downstream pathways such as NF-κB, MAPK, and apoptosis signaling.

As a key mediator of systemic inflammation, TNF-α induces fever, acute-phase protein synthesis, and endothelial activation, while also promoting cytotoxic activity against tumor cells and infected cells. However, its dysregulation is strongly associated with chronic inflammatory and autoimmune diseases, including rheumatoid arthritis, inflammatory bowel disease, psoriasis, and septic shock.

Clinically, TNF-α is a major therapeutic target. Monoclonal antibodies and soluble receptors that neutralize TNF-α are widely used to treat autoimmune conditions. Additionally, TNF-α expression is monitored as a biomarker of inflammation in various disease states, underscoring its dual role in both protective immunity and pathological inflammation.
